Start with Physical Organization

Before calculating, locate information. Is a locus autosomal or X-linked? Are two loci unlinked or on one chromosome? Which alleles share a homologue? A chromosome diagram often resolves what letter strings conceal. Then follow chromosome behavior through meiosis to determine gametes.

State the genotype-to-phenotype rule. Complete dominance, codominance, incomplete dominance, penetrance, polygenic addition and environmental response make different predictions from the same transmission probabilities.

Separate Prediction from Observation

Punnett grids generate expectations. Pedigrees and breeding experiments supply observations. Small differences are normal; systematic departures reveal linkage, viability effects or false assumptions. Chi-squared formalizes comparison but cannot choose among biological explanations without other evidence.

Informative crosses distinguish hypotheses. A recessive tester exposes hidden gametes. An affected daughter challenges simple X-linked recessive inheritance if her father is unaffected. Reciprocal rare classes support crossing over. Large accurately scored samples strengthen every inference.

Genotype Is Not Destiny

Alleles constrain molecular possibilities, while gene interaction, development, environment and chance influence phenotype. PKU treatment, X-inactivation mosaics and temperature-dependent sex determination demonstrate this in different ways. Precision requires avoiding both genetic determinism and the opposite error of ignoring inherited structure.

A complete answer moves from DNA variant to chromosome context, segregation, gamete probability, zygotic genotype, gene-product relationship, phenotype under stated conditions and comparison with evidence. With every link explicit, unfamiliar genetics problems become manageable.
